Nashville took the lead with two goals through its player Jacob Schavelberg, who scored in the 4th and 46th minutes of the match.
Inter Miami was able to reduce the gap with a goal scored by Argentine star Lionel Messi with a pass from Luis Suarez.
Nashville scored a third goal that was not counted after returning to the VAR technique in the 85th minute of the match.
Before the final whistle blew, Luis Suarez scored the equalizer for Inter Miami after a pass from Busquets in the fifth minute of stoppage time.
Inter Miami and Nashville will meet again in the second leg at dawn next Thursday, to decide qualification.
